Overview

Expanded and activated autologous natural killer cells (NKAE cells) is an autologous cell therapy developed by Dr. Joaquín Martínez López and colleagues at Hospital Universitario 12 de Octubre. The therapy involves the ex vivo expansion and activation of a patient's own natural killer (NK) cells, typically derived from peripheral blood, using irradiated feeder cells such as K562-mb15-41BBL or Clone9.mbIL21. These activated cells are then reinfused into the patient to target and eliminate malignant cells. In the context of multiple myeloma, NKAE cells utilize natural cytotoxicity receptors, including NKG2D and NKp30, to recognize and kill myeloma cells. This therapy has been investigated in Phase 1 clinical trials (e.g., NCT02481934) as a consolidation treatment for patients with relapsed or refractory multiple myeloma, often in combination with immunomodulatory drugs like lenalidomide or proteasome inhibitors like bortezomib.
