Overview

Expanded haploidentical NK cells are a form of adoptive cell therapy in which natural killer (NK) cells are collected from a human leukocyte antigen (HLA)-haploidentical donor—typically a parent or close relative—and then expanded and activated ex vivo under good manufacturing practice conditions. The expansion process often involves cytokines such as interleukin-2 or interleukin-21 and feeder cell lines to increase the number and activity of NK cells. These expanded NK cell products are infused into patients, usually after lymphodepleting chemotherapy, with the aim of targeting minimal residual disease or treating malignancies such as high-risk neuroblastoma or leukemia. The mechanism relies on the innate cytotoxicity of allogeneic NK cells against tumor targets via direct killing and antibody-dependent cellular cytotoxicity (ADCC). This approach is being investigated primarily in pediatric oncology settings but has broader potential applications in hematologic malignancies[1][2][3][6].
