Overview

EXS617 is a precision-designed, orally bioavailable, and highly selective small molecule inhibitor of cyclin-dependent kinase 7 (CDK7). Developed using artificial intelligence-driven drug discovery, EXS617 is intended to overcome limitations of existing CDK inhibitors by combining potent, reversible cell cycle and transcription disruption. It is in ongoing Phase 1/2 ("ELUCIDATE") clinical trials for advanced solid tumors, including head and neck cancer, colorectal cancer, pancreatic cancer, non-small cell lung cancer, HR+/HER2- breast carcinoma, and ovarian cancer. Preclinical and early clinical studies have shown strong anti-proliferative activity in multiple tumor types, with a differentiated safety profile, and rational combination strategies with standard-of-care therapies are being explored[1][3][5][9][11][13].
