Drug intelligence / Profile preview
Extended-Release Methylphenidate is an oral, long-acting formulation of the central nervous system (CNS) stimulant methylphenidate. It is widely approved and marketed under various brand names, including Concerta, Ritalin LA, and QuilliChew ER, for the treatment of attention deficit hyperactivity disorder (ADHD) and narcolepsy. The drug acts as a norepinephrine-dopamine reuptake inhibitor (NDRI) by blocking the sodium-dependent dopamine transporter (DAT) and sodium-dependent norepinephrine transporter (NET), thereby increasing synaptic concentrations of these neurotransmitters in the prefrontal cortex and subcortical regions. In addition to its established indications, extended-release methylphenidate is being evaluated by St. Jude Children's Research Hospital in clinical trials (such as the STARS study) to address executive functioning deficits and attention problems in children and adolescents with sickle cell disease.
