Overview

EY-L is an experimental, tumor-targeted liposomal co-formulation combining the mTOR inhibitor everolimus and the survivin inhibitor YM155 (sepantronium bromide). It is specifically designed to enhance the abscopal effect—the regression of distant, non-irradiated tumors following localized treatment—particularly in the context of immunogenic cancers like renal cell carcinoma. When used as part of a trimodal therapy regimen involving radiation and interleukin-2 (IL-2), EY-L works by simultaneously inhibiting the Mammalian target of rapamycin (mTOR) pathway and suppressing the anti-apoptotic protein survivin. This dual action sensitizes tumor cells to radiation-induced damage and promotes immunogenic cell death, thereby facilitating a more robust systemic anti-tumor immune response.
