Overview

Ezrin shRNA is a short hairpin RNA (shRNA) designed to silence the expression of the Ezrin protein (also known as villin-2 or cytovillin). Ezrin is a member of the ERM (Ezrin-Radixin-Moesin) protein family and serves as a cross-linker between the plasma membrane and the actin cytoskeleton, playing a critical role in cell adhesion, migration, and organization of cell surface structures. In the context of oncology, Ezrin is often upregulated and associated with increased metastatic potential. Research, notably from the National Cancer Institute (NCI), has demonstrated that using shRNA to knockdown Ezrin can inhibit metastasis induced by HGF/Met signaling pathways in preclinical melanoma models. This modality works by triggering the RNA interference (RNAi) pathway, leading to the degradation of Ezrin mRNA and a subsequent reduction in protein levels, thereby impairing the invasive capabilities of tumor cells.
