Overview

F12 sildenafil in situ gel is a thermosensitive vaginal formulation of sildenafil citrate developed by researchers at Assiut University. It is specifically designed to treat endometrial thinning, a common adverse effect in women undergoing ovulation induction with clomiphene citrate, particularly those with Polycystic Ovary Syndrome (PCOS). Sildenafil acts as a selective phosphodiesterase type 5 (PDE5) inhibitor, which increases the levels of cyclic guanosine monophosphate (cGMP), leading to smooth muscle relaxation and vasodilation. By enhancing uterine artery blood flow, the drug aims to improve endometrial thickness and receptivity. The F12 formulation is an optimized thermosensitive hydrogel that remains in a liquid state at room temperature for ease of administration and transitions into a bioadhesive gel at body temperature to provide localized, sustained delivery to the uterine tissues while minimizing systemic exposure.
