Overview

F13-PEI1.8k-2 is a fluorinated derivative of branched polyethylenimine (PEI) with a molecular weight of 1.8 kDa, specifically modified with perfluorohexyl (F13) alkyl chains. Developed primarily as a non-viral vector for gene delivery, this polymer addresses the efficiency-toxicity trade-off common in cationic polymers. The incorporation of fluorine atoms leverages the fluorous effect to enhance cellular uptake and facilitate endosomal escape through membrane disruption and the proton sponge effect, while the low-molecular-weight PEI backbone ensures significantly lower cytotoxicity compared to high-molecular-weight variants like PEI 25k. The -2 designation refers to the specific molar ratio of the fluorinated grafting agent to the polymer amines. It is utilized in preclinical research for the delivery of plasmid DNA, siRNA, and mRNA, with potential applications in oncology and genetic disease therapies.
