F16 is a fully human monoclonal antibody, typically utilized in a single-chain variable fragment (scFv) or IgG format, that specifically targets the A1 domain of the large isoform of tenascin-C (TNC). Tenascin-C is an extracellular matrix glycoprotein that is highly overexpressed in the stroma of various solid tumors and during neo-angiogenesis, while remaining virtually absent in normal adult tissues. Developed by Philogen, F16 serves as a targeted delivery vehicle for therapeutic payloads. Its most prominent clinical derivative is **F16-IL2** (also known as **Teleukin**), a recombinant fusion protein consisting of the F16 antibody fused to interleukin-2 (IL-2). This fusion aims to concentrate the immunostimulatory activity of IL-2 within the tumor microenvironment, thereby enhancing anti-tumor immune responses while reducing systemic toxicity. F16 has also been investigated as a fusion with interferon-gamma (F16-IFNgamma) and as a radioimmunotherapeutic agent (e.g., 131I-F16). Clinical trials have explored its efficacy in several malignancies, including non-small cell lung cancer (NSCLC), melanoma, and mesothelioma.
