F17752 is an orally bioavailable small molecule microtubule inhibitor developed by Pierre Fabre Medicament for the treatment of advanced solid tumors. It acts by binding to the colchicine site of tubulin, thereby inhibiting tubulin polymerization and disrupting the mitotic spindle, which leads to cell cycle arrest and apoptosis. In addition to its direct cytotoxic effects, F17752 exhibits vascular disrupting activity (VDA), targeting the established vasculature of tumors to induce rapid necrosis. Clinical development reached Phase I/II, where it was evaluated in a dose-escalation study using a 2-weeks-on/1-week-off schedule, but the program was prematurely terminated.
