F1929-1458 is a small molecule agonist of Nuclear Factor Kappa B Subunit 1 (NFKB1) identified through an in vivo chemical screen. It promotes anti-tumor immunity by activating the NF-κB signaling pathway within tumor cells, which triggers the release of damage-associated molecular patterns (DAMPs) such as ATP, OxLDL, and HMGB1-gDNA complexes. These DAMPs subsequently activate the cGAS-STING and NLRP3 inflammasome pathways in dendritic cells, leading to the production of type I interferons and IL-1β, which facilitate dendritic cell maturation and T-cell-mediated tumor regression. In preclinical models of colorectal cancer, F1929-1458 has demonstrated the ability to remodel the tumor microenvironment and enhance the efficacy of anti-PD-1 immunotherapy.
