**F77 CAR1** is an autologous second-generation **CAR-T cell therapy** being developed for **prostate cancer** at the **University of Pennsylvania**. It is engineered with a single-chain variable fragment derived from an F77-specific mouse monoclonal antibody to recognize the **F77 carbohydrate antigen**, a tumor-associated glycan reported on both androgen-dependent and androgen-independent prostate cancer cells. The construct includes a **CD8 alpha hinge and transmembrane region** with **4-1BB** and **CD3 zeta** intracellular signaling domains, enabling antigen-dependent T-cell activation, cytokine production, and cytotoxic killing of F77-positive tumor cells. In preclinical xenograft models using PC3 prostate cancer cells, F77 CAR1 eradicated tumors, supporting its potential as a targeted cell therapy for prostate cancer and possibly other F77-expressing malignancies.
