F7AK3 is a tetravalent bispecific T-cell engager (TCE) developed by Excyte Biopharma using its proprietary FIST (fusion of IgG and scFv technology) platform. The molecule consists of a humanized anti-TROP2 IgG4P antibody (clone F7A) fused to an anti-CD3 single-chain variable fragment (scFv, clone K3). F7AK3 is designed to treat TROP2-expressing solid tumors, such as triple-negative breast cancer (TNBC), by simultaneously binding to Trophoblast cell-surface antigen 2 (TROP2) on malignant cells and the CD3 receptor on T cells. This dual binding triggers T-cell activation, secretion of cytotoxic cytokines (such as IFN-γ and TNF-α), and directed lysis of the target tumor cells. Preclinical studies have demonstrated its efficacy in inducing potent cytotoxicity in various breast cancer cell lines and inhibiting tumor growth in mouse xenograft models.
