**F8IL10 + methotrexate** is a combination therapy designed for the treatment of active rheumatoid arthritis. F8IL10 (also called Dekavil) is a fusion protein consisting of the F8 single-chain antibody fragment, which targets the extra-domain-A (ED-A) of fibronectin expressed at sites of inflammation, fused to the anti-inflammatory cytokine interleukin-10 (IL10). This construction enables localized delivery of IL10 to inflamed synovial tissue, enhancing anti-inflammatory effects while minimizing systemic exposure. Methotrexate is a standard disease-modifying antirheumatic drug which inhibits dihydrofolate reductase, suppressing immune system overactivity. The combination aims to improve efficacy in rheumatoid arthritis by adding targeted cytokine therapy to conventional immunosuppressive treatment[2][3][1]. Clinical studies have evaluated safety, pharmacokinetics, immunogenicity, and anti-rheumatic activity, focusing on dose escalation and recommended dose finding for the combination[1][2][3].
