Overview

FA-HES-EPI is a pH-responsive, tumor-targeted polymer-drug conjugate designed for the treatment of bladder cancer. It consists of the chemotherapeutic agent epirubicin (EPI) covalently linked to a hydroxyethyl starch (HES) backbone via an acid-sensitive hydrazone bond, with folic acid (FA) moieties attached to provide active targeting to folate receptor-overexpressing tumor cells. The hydroxyethyl starch serves as a biocompatible nanocarrier that improves the solubility and pharmacokinetic profile of epirubicin. Upon internalization into the acidic tumor microenvironment or endosomes, the hydrazone bond cleaves, releasing the active epirubicin payload. This system is often formulated into micelles and has been investigated for the co-delivery of immune modulators like imiquimod to achieve synergistic chemoimmunotherapy.
