Fab14 is a human-specific antigen-binding fragment (Fab) that targets CD68, a glycoprotein highly expressed on the surface of macrophages. Developed by researchers at the University of Alabama at Birmingham and Yale University, Fab14 is designed as an immunoPET imaging probe to noninvasively detect and quantify tumor-associated macrophages (TAMs) within the tumor microenvironment. When radiolabeled with zirconium-89 (as [89Zr]Zr-DFO-Fab14), it allows for the visualization of human macrophage infiltration in humanized mouse models of cancer, such as triple-negative breast cancer. This tool is intended to help study macrophage dynamics and evaluate the efficacy of macrophage-targeted therapies.
