Overview

Fabomotizole is a small molecule anxiolytic drug developed and launched in Russia in the early 2000s, primarily under the brand name Afobazole. It is used for the treatment of anxiety disorders, neurasthenia, adjustment disorders, sleep disturbances related to anxiety, and other stress-related conditions. Unlike benzodiazepines, it does not cause sedation or muscle relaxation and has a low risk of dependence. Its mechanism of action is not fully defined but includes modulation of sigma-1 receptor (Sigma1R), reversible inhibition of monoamine oxidase A (MAO-A), agonism at melatonin MT1 receptor, antagonism at MT3 receptor (NQO2), promotion of nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF) release, as well as possible effects on GABAergic systems and serotonin receptors. Fabomotizole has demonstrated neuroprotective properties in preclinical studies and is reported to be well tolerated with few side effects[1][5][6][8].
