Anti-FABP4 neutralizing antibody is a research-stage monoclonal antibody therapeutic designed to target and neutralize circulating fatty acid binding protein 4 (FABP4). FABP4 is an adipokine primarily secreted by adipocytes that plays a critical role in metabolic homeostasis and the progression of certain malignancies. By binding to and neutralizing FABP4, the antibody blocks its interaction with the ITGB1 receptor, subsequently inhibiting the PI3K/AKT/β-catenin signaling cascade. Preclinical studies, including research from The Hong Kong Polytechnic University presented at AACR 2024, have demonstrated that humanized versions of this antibody can reduce hepatic steatosis in models of metabolism-associated steatotic liver disease (MASLD) and suppress the development of NAFLD-induced hepatocellular carcinoma (HCC) by inhibiting cancer stem cell functions. It is also under investigation for its potential efficacy in treating breast cancer.
