FACT-222 is an investigational allogeneic chimeric antigen receptor (CAR) T-cell therapy designed to target the CD22 antigen, a surface protein commonly expressed on B-cell malignancies. Developed by Factor Bioscience, the therapy utilizes proprietary mRNA-based cell engineering technology (TALEN) to create "off-the-shelf" CAR-T cells from healthy donors, potentially overcoming the logistical and scalability challenges associated with autologous therapies. The primary indication for FACT-222 is acute lymphoblastic leukemia (ALL), specifically for patients who have relapsed or are refractory to existing treatments, including CD19-targeted CAR-T therapies. By targeting an alternative B-cell antigen (CD22), FACT-222 aims to address antigen escape and provide a therapeutic option for patients with high unmet clinical needs. The asset was part of a portfolio being acquired by Tempest Therapeutics to expand its cell therapy pipeline.
