Overview

FADS1-shRNA is a short hairpin RNA (shRNA) construct designed to silence the expression of Fatty acid desaturase 1 (FADS1). FADS1 is a key rate-limiting enzyme in the metabolism of omega-3 and omega-6 polyunsaturated fatty acids (PUFAs). In preclinical studies involving renal cell carcinoma (RCC) cell lines, the use of FADS1-shRNA to knock down FADS1 expression resulted in significantly reduced cell proliferation, downregulation of DNA biosynthesis and cell division pathways, and altered purine metabolism. The anti-proliferative effect is potentially mediated through the inhibition of the MAPK pathway, specifically evidenced by reduced P38 phosphorylation. This research identifies FADS1 as a potential prognostic marker and therapeutic target in various cancers, particularly RCC.
