FAKnostics is developing a hydrocarbon-stapled peptide that specifically targets the Focal Adhesion Targeting (FAT) domain of Focal Adhesion Kinase (FAK, also known as PTK2). Unlike traditional small-molecule FAK inhibitors that target the ATP-binding kinase domain, this peptide is designed to disrupt the non-kinase scaffolding functions of FAK. By interfering with the protein-protein interactions mediated by the FAT domain (such as those with paxillin and VEGFR3), the drug aims to inhibit tumor cell survival, migration, and the development of the dense, fibrotic stroma characteristic of pancreatic ductal adenocarcinoma (PDAC). The program is currently in the IND-enabling phase of development.
