Overview
This is a direct-acting antiviral **triple drug combination** composed of faldaprevir (an NS3/4A protease inhibitor), TD-6450 (an NS5A inhibitor), and ribavirin (a nucleoside analogue). The regimen was developed and clinically tested by Trek Therapeutics for the treatment of chronic hepatitis C virus (HCV) infection, specifically in genotypes 1b and 4. Faldaprevir directly inhibits the NS3/4A protease, essential for viral replication, while TD-6450 blocks the NS5A protein required for both viral RNA replication and assembly, and ribavirin acts as a broad-spectrum antiviral likely through inhibition of inosine monophosphate dehydrogenase and RNA mutagenesis. Clinical trials demonstrated high sustained virologic response (SVR) rates in treated patients with good tolerability[1][2][3][4][6].
