Overview

Famciclovir is a second-generation oral nucleoside analogue antiviral drug primarily used to treat infections caused by herpesviruses, including herpes zoster (shingles), genital herpes (herpes simplex virus type 2), and cold sores (herpes labialis). It is also indicated for the treatment of recurrent mucocutaneous herpes simplex in HIV-infected patients. Famciclovir is a prodrug that is rapidly converted in the liver and intestinal wall to penciclovir, its active form. Penciclovir acts as a competitive inhibitor of viral DNA polymerase after being phosphorylated by viral thymidine kinase within infected cells. This inhibits viral DNA synthesis and replication without affecting normal cellular processes. Famciclovir does not cure herpes infections but reduces symptoms and frequency of outbreaks[1][5][4].
