Famitinib + SHR-A1811 is an investigational combination therapy being evaluated for the treatment of advanced breast cancer, particularly HR-positive/HER2-negative cases that have developed resistance to CDK4/6 inhibitors. Famitinib is a small-molecule multi-kinase inhibitor developed by Jiangsu Hengrui Pharmaceuticals that targets VEGFR2, PDGFR, and c-Kit, thereby inhibiting angiogenesis and tumor proliferation. SHR-A1811 is a next-generation antibody-drug conjugate (ADC) also developed by Jiangsu Hengrui Pharmaceuticals, consisting of a humanized anti-HER2 monoclonal antibody linked to a topoisomerase I inhibitor payload (SHR152852) via a cleavable linker. The combination is currently being studied in a Phase 3 trial led by Fudan University, focusing on the SNF4 molecular subtype of breast cancer to determine if the dual inhibition of angiogenic pathways and HER2-mediated signaling can improve outcomes in treatment-resistant populations.
