Overview

Famprofazone is a nonsteroidal anti-inflammatory drug (NSAID) of the pyrazolone series with analgesic, anti-inflammatory, and antipyretic effects. It is available over-the-counter in some countries such as Taiwan and is found in multi-ingredient medications like Gewodin and Gewolen. Famprofazone acts primarily by inhibiting cyclooxygenase (COX), thereby reducing prostaglandin synthesis responsible for pain, inflammation, and fever. A notable pharmacological feature of famprofazone is its metabolism to methamphetamine and amphetamine; approximately 15–20% of an oral dose can be converted to methamphetamine. This metabolic pathway has led to positive drug tests for amphetamines in users. Due to safety concerns related to this metabolism, its use has been discontinued or restricted in many countries[1][2][3][4][5].
