**FAP-CAR-ΔTGFβRII** is a preclinical, mRNA-encoded CAR-macrophage construct designed to target fibroblast activation protein alpha on cancer-associated fibroblasts while co-expressing a dominant-negative transforming growth factor beta receptor 2 module. FAP-directed CAR activity is intended to enable macrophage-mediated elimination of fibroblast activation protein alpha-positive stromal cells, whereas the truncated transforming growth factor beta receptor 2 blocks immunosuppressive transforming growth factor beta signaling and reduces profibrotic activity. The construct was developed as one component of an intravenously delivered lipid nanoparticle mRNA system that generated in vivo CAR-macrophages in mouse hepatocellular carcinoma models; it was co-administered with GPC3-CAR-Super IL-2 in the quadrivalent LNP-GF CAR regimen.
