Overview

FAP-GPC3 bispecific CAR-T cell therapy is an investigational cell-based gene therapy developed by the Guangzhou Institutes of Biomedicine and Health (GIBH), Chinese Academy of Sciences, in collaboration with Guangdong Zhaotai Cell Biology Technology Ltd. This therapy is specifically designed to treat hepatocellular carcinoma (HCC) by simultaneously targeting two distinct antigens: Fibroblast Activation Protein (FAP) and Glypican 3 (GPC3). FAP is typically expressed on cancer-associated fibroblasts and some tumor cells, while GPC3 is a well-known marker frequently overexpressed in HCC. By utilizing a bispecific chimeric antigen receptor (CAR), the engineered T cells can recognize and be activated by either antigen, a strategy intended to overcome tumor heterogeneity and prevent antigen escape—common mechanisms of resistance to single-target CAR-T therapies. Preclinical studies have demonstrated that these bispecific CAR-T cells exhibit more robust antitumor activity and significantly prolonged survival in mouse models compared to single-target anti-FAP or anti-GPC3 CAR-T cells.
