FAP-Lu177 is an investigational targeted radiopharmaceutical developed by Ratio Therapeutics. It consists of a lutetium-177 (Lu-177) radioisotope conjugated to a ligand that specifically targets Fibroblast Activation Protein (FAP). FAP is highly expressed in the tumor stroma, particularly in cancer-associated fibroblasts, across a wide range of solid tumors, while maintaining low expression in most normal tissues. By binding to FAP, the drug delivers localized beta-particle radiation to the tumor microenvironment, aiming to induce DNA damage and cell death in both the stroma and adjacent tumor cells. The program is currently in Phase 1 clinical development, including evaluation through investigator-initiated trials and compassionate use programs for patients with multiple solid tumor types.
