The FAP mRNA vaccine is a therapeutic cancer vaccine developed by West China Hospital, currently in Phase I clinical development for the treatment of advanced malignant solid tumors. The vaccine utilizes messenger RNA (mRNA) technology to induce an immune response against Fibroblast Activation Protein (FAP), a cell-surface glycoprotein that is highly and selectively expressed on cancer-associated fibroblasts (CAFs) within the tumor microenvironment. CAFs play a critical role in tumor progression by creating physical and functional barriers that limit the infiltration of immune cells and the delivery of therapeutic agents. By targeting and depleting FAP-positive CAFs, the vaccine aims to remodel the tumor stroma, thereby disrupting the immunosuppressive environment and enhancing the efficacy of concurrent therapies, such as immune checkpoint inhibitors. The vaccine is being evaluated in a basket study to assess its safety, tolerability, and preliminary anti-tumor activity.
