FAP mRNA vaccine + immune checkpoint inhibitors is a combination immunotherapy regimen being developed by West China Hospital for the treatment of advanced malignant solid tumors. The regimen consists of a therapeutic mRNA vaccine that targets Fibroblast Activation Protein (FAP), a marker highly expressed on cancer-associated fibroblasts (CAFs), and immune checkpoint inhibitors (ICIs). CAFs contribute to an immunosuppressive tumor microenvironment and act as a physical barrier to therapy. By targeting FAP, the vaccine aims to induce an immune response that depletes CAFs and remodels the tumor stroma, thereby improving the infiltration of immune cells and the delivery of therapeutic agents. The addition of immune checkpoint inhibitors further enhances the anti-tumor immune response by overcoming checkpoint-mediated immunosuppression. This combination is currently being evaluated in a Phase I exploratory basket study.
