Overview

FAP-specific Affilin is an engineered non-immunoglobulin scaffold protein based on a ubiquitin mutein (Affilin) that binds fibroblast activation protein (FAP) with high affinity and specificity for use as a targeting ligand in diagnostic and therapeutic applications, particularly in oncology. Affilin molecules from Navigo Proteins are designed as small, stable, easily expressed proteins that can be modularly fused or conjugated to radioisotopes, cytotoxic drugs, or other effector payloads, enabling FAP-targeted radiopharmaceuticals and other precision therapies aimed at cancer-associated fibroblasts and FAP-expressing stromal cells in solid tumors.[1][2] FAP-specific Affilin constructs show strong binding to human and mouse FAP, maintain affinity after prolonged serum incubation, and are being developed in collaboration with ITM Isotope Technologies Munich as the targeting component of FAP-directed radionuclide therapies for a broad range of solid tumors.[1][2]
