Overview

FAP-targeted immunosuppressive CAR-DC is an autologous cell therapy being developed by the Second Affiliated Hospital, School of Medicine, Zhejiang University for the treatment of end-stage idiopathic pulmonary fibrosis (IPF). The therapy utilizes chimeric antigen receptor (CAR) technology applied to dendritic cells (DCs) rather than T cells. These engineered immunosuppressive CAR-DCs (iCAR-DCs) are designed to specifically target fibroblast activation protein (FAP), which is highly expressed on pathological CTHRC1+ fibroblasts—the primary drivers of collagen production and irreversible scarring in IPF. By localizing to fibrotic lung areas, these cells aim to attenuate fibrosis and improve patient survival without eliciting detrimental immune responses or systemic toxicity.
