Overview

Farampator is a small molecule drug belonging to the class of ampakines, which act as positive allosteric modulators of AMPA-type glutamate receptors. By binding to these receptors, farampator enhances excitatory neurotransmission mediated by glutamate, leading to increased synaptic plasticity and improved cognitive function. It was primarily investigated for its potential therapeutic effects in cognitive disorders such as Alzheimer's disease and schizophrenia, with additional research in major depressive disorder. Farampator was developed by Cortex Pharmaceuticals and licensed to Organon BioSciences for commercial development; after Organon's acquisition by Schering-Plough, the license transferred accordingly. Clinical studies showed that farampator could improve short-term memory but might impair episodic memory and cause side effects like headache, somnolence, and nausea—particularly at higher plasma levels. Development was ultimately terminated due to concerns about cardiac toxicity[1][3][4][6].
