Faricizumab is a bispecific monoclonal antibody designed for the treatment of retinal vascular diseases. It is the first bispecific antibody approved for ophthalmic use, engineered to simultaneously bind and neutralize two distinct pathways: vascular endothelial growth factor-A (VEGF-A) and angiopoietin-2 (Ang-2). By inhibiting VEGF-A, faricizumab reduces vascular permeability and angiogenesis, while its inhibition of Ang-2 promotes vascular stability and desensitizes the blood vessels to the effects of VEGF-A. Developed by Roche and its subsidiary Genentech, faricizumab is administered via intravitreal injection. It is currently approved for the treatment of neovascular (wet) age-related macular degeneration (nAMD), diabetic macular edema (DME), and retinal vein occlusion (RVO). It is also under investigation for proliferative diabetic retinopathy and other retinal conditions.
