Overview

Farletuzumab ecteribulin is an investigational **folate receptor alpha-targeting antibody-drug conjugate** developed by Eisai. It comprises the humanized IgG1 anti-folate receptor alpha antibody farletuzumab conjugated through an enzymatically cleavable, cathepsin B-cleavable linker to the microtubule-dynamics inhibitor eribulin. Following binding to folate receptor alpha-positive tumor cells and internalization, linker cleavage releases eribulin, which disrupts microtubule dynamics and produces cytotoxic antitumor activity; released payload may also contribute to a bystander effect. The agent has been evaluated in ovarian, endometrial, non-small cell lung, primary peritoneal, fallopian tube, and triple-negative breast cancers. Eisai regained sole global development and commercialization rights after its collaboration with Bristol Myers Squibb ended on July 1, 2024.
