Fas-4-1BB CAR T cells are an experimental adoptive cell therapy modality where T cells are engineered to express both a Chimeric Antigen Receptor (CAR) and a Fas-4-1BB immunomodulatory fusion protein (IFP). The IFP consists of the extracellular and transmembrane domains of the Fas death receptor (CD95) fused to the intracellular signaling domain of the 4-1BB (CD137) co-stimulatory receptor. In the tumor microenvironment, where Fas ligand (FasL) is often upregulated to induce T cell apoptosis, this fusion protein acts as a "signal converter," transforming the pro-apoptotic FasL signal into a pro-survival and proliferative 4-1BB signal. This approach aims to overcome tumor-induced immunosuppression, enhancing the persistence, metabolic fitness, and anti-tumor activity of CAR T cells in both solid and hematologic malignancies.
