FAS93 is a potent, small-molecule inhibitor of **Fatty Acid Synthase (FASN)**, a multi-enzyme protein that plays a central role in de novo lipogenesis by converting acetyl-CoA and malonyl-CoA into long-chain fatty acids. Developed by **3-V Biosciences** (now **Sagimet Biosciences**), FAS93 specifically targets the **ketoacyl reductase (KR) domain** of the FASN complex. By blocking the production of palmitate, FAS93 disrupts the formation of lipid rafts and the metabolic pathways required for viral replication and the survival of lipogenic cancer cells. Preclinical studies have demonstrated its efficacy against a range of viruses, including **Hepatitis C virus (HCV)** and **Rhinovirus**, as well as various solid tumors. Although FAS93 was a key early compound in the 3-V Biosciences portfolio, the company has primarily advanced other FASN inhibitors, such as denifanstat (TVB-2640), into clinical development for indications like metabolic-associated steatohepatitis (MASH) and oncology.
