FATE-NK100 is a first-in-class, allogeneic, donor-derived natural killer (NK) cell cancer immunotherapy comprised of adaptive memory NK cells. These cells are sourced from healthy donors and enriched for an "adaptive" phenotype, often associated with prior human cytomegalovirus (CMV) exposure. The product is manufactured through a feeder-free, seven-day ex vivo activation process using pharmacologic modulators to enhance anti-tumor activity. Mechanistically, FATE-NK100 is designed to directly kill tumor cells, resist immune checkpoint inhibition, and promote antibody-dependent cellular cytotoxicity (ADCC). It has demonstrated enhanced cytokine production and persistence compared to conventional NK cell therapies. Developed by Fate Therapeutics in collaboration with the University of Minnesota and Masonic Cancer Center, its primary indications included acute myeloid leukemia (AML), ovarian cancer, fallopian tube cancer, peritoneal cancer, solid tumors including breast and colorectal cancers—both as monotherapy and in combination with monoclonal antibodies such as trastuzumab or cetuximab[2][3][4][5][6][10].
