Overview

The combination of **favezelimab**, **pembrolizumab**, and **vibostolimab** brings together three different monoclonal antibodies with distinct but complementary immunomodulatory mechanisms. - **Pembrolizumab** is a humanized monoclonal antibody targeting programmed death-1 (**PD-1**) to block an immune checkpoint pathway and allow T-cell–mediated antitumor response. - **Favezelimab** targets lymphocyte-activation gene 3 (**LAG-3**), another inhibitory receptor on T cells, with the intention of further reversing T-cell exhaustion and promoting antitumor immunity. - **Vibostolimab** is an anti–TIGIT antibody, inhibiting the **T-cell immunoreceptor with Ig and ITIM domains (TIGIT)**, a checkpoint also associated with tumor immune evasion. This triple combination has primarily been developed and investigated by **Merck** in the context of advanced or refractory cancers, particularly non-small cell lung cancer (NSCLC) and classical Hodgkin lymphoma, targeting patients who have progressed on prior therapies. Despite early investigations, late-stage trials involving these drugs—both in double and triple combinations—have been discontinued following lack of efficacy, particularly in NSCLC, and disappointing results in clinical development programs.
