Overview

FAZ053 + PDR001 is a combination therapy under investigation for advanced solid tumors and other malignancies. FAZ053 is a monoclonal antibody targeting programmed death-ligand 1 (PD-L1), which blocks the interaction between PD-L1 on tumor cells and PD-1 on T cells, thereby releasing the inhibition of T cell activity against tumors. PDR001 (also known as spartalizumab) is an anti-PD-1 monoclonal antibody that suppresses the programmed death-1 pathway, further promoting immune-mediated recognition and destruction of cancer cells. Both are immune checkpoint inhibitors designed to enhance antitumor immune responses. The combination was studied in Phase I trials for advanced cancers, including alveolar soft part sarcoma, chordoma, colon cancer, and triple-negative breast cancer. The anticipated synergistic benefit of co-blockade of PD-1 and PD-L1 was not clearly observed, but both agents individually and in combination were generally well tolerated[1][3][5][7].
