Overview

FB520 is a small molecule therapeutic candidate being developed by 1ST Biotherapeutics for the treatment of oncology. It targets the Integrated Stress Response (ISR) pathway by inhibiting three key kinases: General Control Nondenressible 2 (GCN2), Protein Kinase RNA-like Endoplasmic Reticulum Kinase (PERK), and Heme-Regulated Inhibitor (HRI). These kinases are activated in response to various cellular stresses, such as nutrient deprivation, hypoxia, or proteotoxic stress, which are common in the tumor microenvironment. By inhibiting these stress-response kinases, FB520 aims to disrupt the survival mechanisms of cancer cells, prevent the translation of stress-adaptive proteins, and promote apoptosis. It is currently in the discovery phase of development.
