FBL-MTX is a novel formulation of methotrexate in which the active drug is encapsulated within folate-based liposomes. This design aims to enhance targeted delivery and reduce systemic toxicity by exploiting the affinity of folic acid for certain cell types, such as activated macrophages and inflamed tissues seen in rheumatoid arthritis. Methotrexate itself is an antimetabolite and antifolate agent that inhibits dihydrofolate reductase (DHFR), thereby blocking DNA synthesis and exerting immunosuppressive effects. The folate-liposome formulation may improve patient convenience (notably via subcutaneous administration) and potentially increase efficacy or tolerability compared to conventional methotrexate therapy. FBL-MTX is being developed primarily for the treatment of rheumatoid arthritis[1][6].
