FBM5712 is an investigational small‑molecule inhibitor of activin receptor‑like kinase 5 (ALK5), the kinase domain of the transforming growth factor beta receptor 1 (TGF‑βR1), being developed by Thirona Bio for the local treatment of fibrotic diseases.[2][3][9][12] By selectively blocking ALK5/TGF‑β signaling, FBM5712 is designed to reduce pathologic collagen deposition and tissue scarring while limiting systemic exposure and toxicity.[2][3][9][12] Thirona is advancing FBM5712 primarily as a topical therapy for skin fibrosis in conditions such as scleroderma, keloid scars, and certain cancers, while MannKind is collaborating to develop MNKD‑501, an inhaled dry‑powder formulation of FBM5712 for fibrotic lung diseases including idiopathic pulmonary fibrosis.[1][2][3][9][10][11]
