Drug intelligence / Profile preview
FcγRIIA blocking monoclonal antibodies represent a class of therapeutic and research-grade antibodies designed to target and inhibit FcγRIIA (CD32a), a low-affinity activating IgG Fc receptor expressed on platelets, neutrophils, monocytes, and macrophages. By blocking the interaction between IgG immune complexes and FcγRIIA, these antibodies prevent immune-mediated cellular activation, platelet aggregation, and subsequent clearance or thrombosis. This mechanism is highly relevant in the treatment of immune-mediated thrombocytopenias, such as immune thrombocytopenia (ITP), heparin-induced thrombocytopenia (HIT), and vaccine-induced immune thrombotic thrombocytopenia (VITT), as well as other immune complex-mediated autoimmune diseases like systemic lupus erythematosus (SLE) and ANCA-associated vasculitis. While the murine clone IV.3 is widely utilized as a research tool to study FcγRIIA-dependent pathways, humanized and effector-deficient variants (such as VIB9600/MEDI9600 and the monovalent sc9600) have been developed for clinical evaluation to avoid the Fc-mediated platelet clearance and anaphylaxis associated with wild-type divalent antibodies.
