Drug intelligence / Profile preview
Fc-CLEC-2 (also known as CLEC-2-Fc or CLEC2.Fc) is a recombinant fusion protein consisting of the extracellular domain of C-type lectin-like receptor 2 (CLEC-2, encoded by the CLEC1B gene) fused to an immunoglobulin Fc domain (typically human IgG1 Fc). It acts as a soluble decoy receptor or agonist depending on the context. Fc-CLEC-2 physically binds to podoplanin (PDPN), the endogenous ligand of CLEC-2, which is expressed on various cell types including kidney podocytes, lymphatic endothelial cells, and inflammatory macrophages. In research settings, Fc-CLEC-2 is used to study podoplanin-CLEC-2 interactions, podocyte pathobiology, and macrophage migration. Additionally, it has been investigated preclinically as a therapeutic candidate for several indications: by Amgen for diabetes and liver disease (Kupffer cell polarization), by the National Health Research Institute (NHRI) in Taiwan for SARS-CoV-2-mediated thromboinflammation, and by the University of Birmingham for peritonitis and tissue inflammation.
