Fc-MDNA132 is an engineered interleukin-13 (IL-13) superkine fused to an Fc scaffold to enhance its pharmacokinetic profile and extend its half-life. Developed by Medicenna Therapeutics, it is designed to selectively target the IL-13 decoy receptor (IL13Rα2), which is highly overexpressed in various solid tumors including glioblastoma, pancreatic, colorectal, and prostate cancers, while exhibiting minimal binding to the functional Type II IL-4 receptor (IL4Rα/IL13Rα1) found on normal tissues. This high specificity allows Fc-MDNA132 to serve as a targeted delivery vehicle for therapeutic immune modulators directly to the tumor microenvironment. By binding to IL13Rα2 without being rapidly internalized, it facilitates durable exposure of attached immune-modulatory payloads, such as IL-2 agonists or checkpoint inhibitors, at the tumor site.
