Overview

Fc-mox40l is a recombinant fusion protein composed of the extracellular domain of murine OX40 ligand (OX40L, CD252, TNFSF4) linked to the Fc region of immunoglobulin G. It acts as a potent agonist of the OX40 receptor (CD134, TNFRSF4), a member of the tumor necrosis factor receptor superfamily present on activated T cells. By engaging OX40 on T cells, fc-mox40l provides a co-stimulatory signal that enhances both CD4+ and CD8+ T cell proliferation, survival, and cytokine production, while inhibiting suppressive regulatory T cells. In murine models of colon carcinoma and renal cell carcinoma, fc-mox40l demonstrated superior anti-tumor activity and produced complete remissions compared to OX40 agonist antibody controls, also promoting long-lasting survival and reshaping the tumor immune microenvironment[1][3][4]. The Fc fusion is designed for improved stability and extended half-life compared to OX40L alone. This agent is used primarily in preclinical and translational cancer immunotherapy research.
