Fc-Saxatilin is a recombinant Fc-fusion protein based on saxatilin, a snake venom disintegrin that contains an Arg-Gly-Asp (RGD) motif and exerts potent thrombolytic and antiplatelet effects by antagonizing multiple integrins on platelets and endothelial cells, including αIIbβ3, α5β1, αvβ3, αvβ1, and αvβ5.[9][11][10] By fusing saxatilin to the Fc portion of immunoglobulin, Fc-Saxatilin extends the plasma half-life and is being developed as an integrin antagonist for the treatment of ischaemic stroke, where it both promotes thrombus resolution and prevents VEGF- or hypoxia-induced vascular leakage by regulating tight-junction proteins such as claudin-5 and occludin and inhibiting Src and Rac1 activation in brain microvascular endothelial cells.[1][7][11][2] The program was initially developed at Yonsei University and is being co-developed with Celltrion, with nonclinical and early clinical work focused on nervous system indications involving cerebral ischaemia and blood–brain barrier injury.[1][5][7][11]
