Fc-silenced anti-CD47 (mIAP301 LALA) is a recombinant, Fc-silent monoclonal antibody designed for preclinical research, particularly in mouse models of hematopoietic stem cell transplantation (HSCT). It is derived from the anti-mouse CD47 clone mIAP301, which was originally a mouse IgG2a isotype. The antibody has been engineered with LALA (L234A, L235A) mutations in the Fc region to abolish Fc-mediated effector functions such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). By targeting CD47 (integrin-associated protein), the antibody blocks the "don't eat me" signal, facilitating the depletion of hematopoietic stem cells (HSCs) as part of a non-genotoxic conditioning regimen. The Fc-silencing modification is specifically intended to mitigate the severe anemia typically associated with CD47 blockade on red blood cells, while maintaining the efficacy of HSC clearance. This compound is primarily investigated for conditioning in the treatment of non-malignant hematologic diseases like sickle cell disease.
