Drug intelligence / Profile preview
FcgRIIb agonist antibody refers to a class of therapeutic monoclonal antibodies designed to selectively target and activate Fc gamma receptor IIb (FcgRIIb, also known as CD32B), the sole inhibitory receptor within the Fc gamma receptor family [vkag141.035]. Co-ligation of FcgRIIb with activating receptors triggers inhibitory signaling (such as SHIP-1 recruitment) that downregulates immune cell activation [vkag141.035]. Developing selective agonists has historically been challenging due to the 96% sequence identity between the extracellular domains of FcgRIIb and the activating receptor FcgRIIa [vkag141.035]. Seismic Therapeutic has utilized its machine learning-enabled IMPACT platform to discover selective FcgRIIb agonist antibodies that avoid activating FcgRIIa, demonstrating the ability to reduce antigen-specific antibody responses in preclinical models of autoimmune and inflammatory diseases [vkag141.035].
